Аннотация: Homoserine Lactones (HSLs) and Resorcinolic Lipids (RLs) originating from microbes and plants influence immune system, however, it is currently unclear whether HSLs and RLs are activators or suppressors of the cytokine network, especially in humans. A study has been carried out to investigate the effects of pure, synthetic HSLs and RLs with different chain lengths on pro-and anti-inflammatory cytokines produced by human blood monocyte cultures. Human monocytes (macrophages and lymphocytes) were collected from leukocyte-rich plasma, separated on a double density gradient, pre-treated with HSLs or Rls and then were inducted with standard either lipopolysaccharide or phytohemagglutinin stimulus. Quantitative assays for pro-and anti-inflammatory cytokines in the monocyte culture supernatants were performed using enzyme immunoassay kits for the quantitative determination of IL-1β, IL-2, IL-4, IL-10, TNF-α and IF-γ. We observed differences in cytokine production according to HSL and RL pre-treatment, increasing in the following order: IL-1β→IL-2≈IL-10≈TNF-α→IL-4→IF-γ. HSLs have been shown more pronounced inhibitory effects than Rls and long-chained homologs were more active than short-chained ones. Our study suggests that some small molecules originating from microbes and plants play an important role in cytokine network regulation and the immunomodulatory effect of HSLs and RLs may obstruct host defenses and affect inflammatory processes

Аннотация: The immune status of basically healthy personnel of OAO Uralkalii and OAO Sil'vinit was studied for three years (1998-2000) to assess the effect of the workplace environment on the parameters of the immune system. All probands were divided into groups depending on their exposure to salts or halite waste. Annual examination did not reveal any statistically significant differences between the parameters of immunograms in the groups studied. The parameters of the humoral component of the immune system of potassium-industry workers manifested a stable tendency toward normalization, irrespective of whether or not these persons were exposed to salts, and the annual contents of T and B lymphocytes and phagocytizing cells were stable within the regional standards

Аннотация: The effect of native а-fetoprotein (AFP) on the expression of T-regulatory lymphocyte (Treg) markers by activated CD4+ lymphocytes with different proliferative status was studied. а-Fetoprotein did not affect the ratio of proliferating and non-proliferating activated CD4+ cells. In the study of Treg differentiation, it was found that AFP at concentrations of 50 and 100 mg/mL significantly inhibited the number of non-proliferating CD4+FOXP3+ and CD4+FOXP3+HELIOS+ lymphocytes without affecting the expression of Treg markers by proliferating CD4+ lymphocytes.

Аннотация: High prevalence of non-AIDS-defining illnesses in treated HIV-infected patients is associated with increased peripheral CD8.sup.+ T cell counts. Hepatitis C virus (HCV) coinfection is an additional risk factor for the development of non-AIDS events. We found that, in HIV/HCV coinfection, the increased proportion of CD8.sup.+ T lymphocytes is due to the effector memory and terminal effector T cells gain. Moreover, in these patients, the accumulation of highly differentiated forms of CD8.sup.+ T lymphocytes was associated with increased concentrations of inflammatory indices.

Аннотация: The effect on activity of peripheral blood lymphocytes upon blast-transformation reaction induced by phytohemagglutinin (PGA) in different concentrations was investigated. For the first time the different-directed activity of MP-6 tetrapeptide in respect to stimulated PGA and non-stimulated peripheral blood lymphocytes is shown. Dependence of MP-6 effects on its own concentration as well as on the mitogen concentration in the cell culture is demonstrated. Myelopid and MP-5 didn't affect the lymphocyte functional activity

Аннотация: Glycolipid biosurfactant (GLB) from Rhodococcus ruber IEGM 231 was found to stimulate tumor necrosis factor-α (TNF-α), interleukin (IL) -1β and IL-6 production when applied as an ultrasonic emulsion to the adherent human peripheral blood monocyte culture. However, a lack of cytokine-stimulating activity was registered with the GLB applied as a hydrophobic film coating in 24-well culture plates, indicating that it may have been due to its inhibitory effect on monocyte adhesion. The mode of GLB application may therefore play an important role in in vitro assay of immunostimulatory activity of this compound as well as other bacterial glycolipids. Аннотация: This paper proposes a quantitative model of the reaction-diffusion type to examine the distribution of interferon-alpha (IFN alpha) in a lymph node (LN). The numerical treatment of the model is based on using an original unstructured mesh generation software Ani3D and nonlinear finite volume method for diffusion equations. The study results in suggestion that due to the variations in hydraulic conductivity of various zones of the secondary lymphoid organs the spatial stationary distribution of IFN alpha is essentially heterogeneous across the organs. Highly protected domains such as sinuses, conduits, co-exist with the regions in which where the stationary concentration of IFN alpha is lower by about 100-fold. This is the first study where the spatial distribution of soluble immune factors in secondary lymphoid organs is modelled for a realistic three-dimensional geometry
