Аннотация: The diastereoselective acylation of a number of racemic methyl-substituted cyclic alkylamines with active esters of 2-phenoxypropanoic acid was studied in detail. The ester of (R)-2-phenoxypropanoic acid and N-hydroxysuccinimide was found to be the most selective agent. The highest stereoselectivity was observed in the kinetic resolution of racemic 2-methylpiperidine in toluene at −40 °C (selectivity factor s = 73) with the predominant formation of (R,R)-amide (93.7% de). To explain the observed stereoselectivity, DFT modelling of the transition states in the reactions of the title acylating agent with 2-methylpiperidine and 2-methylpyrrolidine was performed. The calculated values were in good agreement with experimental data. It has been demonstrated that the acylation proceeds via a concerted mechanism, in which the addition of an amine occurs simultaneously with the elimination of the hydroxysuccinimide fragment. The high stereoselectivity of the (R,R)-amide formation is largely ensured by the lower steric hindrances in the transition states as compared to the formation of (R,S)-amide.

Аннотация: Condensation of fluorinated 3-oxo esters or 1,3-diketones with benzaldehyde and (thio)urea results in the diastereoselective formation of 4-fluoroalkyl-4-hydroxy-2-oxo(thioxo)-6-phenyl-hexahydropyrimidine-5-carboxylates from which by dehydration under acidic conditions the corresponding 6-fluoroalkyl-2-oxo(thioxo)-4-phenyl-1,2,3,4-tetrahydropyrimidine-5-carboxylates were obtained. Under the same conditions, hexafluoroacetylacetone furnishes 4,6-dihydroxy-4,6-di(trifluoromethyl)-hexahydropyrimidin-2-one. Some further reactions of these pyrimidine derivatives leading to fused heterocycles are described.????

Аннотация: Two ethyleneglycol esters of 1-pyrene carboxylic acid were studied as chemosensors for the fluorescence “turn-off” detection of two nitro-containing analytes, such as antiviral drug Riamilovir (Triazavirin ®) and herbicidal agent dinitro-ortho-cresol (DNOC). In both cases the dramatic fluorescence quenching was observed with quenching constants as high as 3·104 M-1 and limits of detection (LOD) as low as 100 ppb.

Аннотация: An efficient approach for the synthesis of 3-(polyfluoroacyl)pyruvaldehydes dimethyl acetals (1,1-dimethoxy-4-polyfluoroalkyl-butan-2,4-dions) from 1,1-dimethoxyacetone and polyfluorinated carboxylic acid esters has been developed. The procedure includes the Claisen type condensation of the starting materials by means of calcium hydride in methanol, followed by isolation of copper complexes of the corresponding diketones and their destroying with disodium EDTA. A simple synthesis of 5(3)-(polyfluoroalkyl)-1H-pyrazole-3(5)-carbaldehydes and their acetals is also presented.

Аннотация: A large laboratory scale (ca. 40 g) method for the preparation of enantiomerically pure (S)- and (R)-valine tert-butyl esters has been developed. The method involves three steps: preparation of N-TFA-valines, preparation of valine tert-butyl esters using 2-methylpropene in dioxane in the presence of sulfuric acid, and isolation of the target compounds as the acetate derivative. The overall yield is up to 70% relative to the starting valine, ee being more than 98% (by HPLC)????

Аннотация: The structure of key intermediates in the synthesis of fluoroquinolone antibiotics: diethyl (2,3,4,5-tetrafluorobenzoyl)malonate and ethyl (2Z)-3-hydroxy-(2,3,4,5-tetrafluorophenyl)prop-2-enoate was for first time studied using X-ray diffraction (XRD) and H-1, F-19 NMR spectroscopy. In solution both the esters were shown to exist as a mixture of enol and ketone tautomeric forms with predominance of the latter. Аннотация: The reactions of ethyl 2-ethoxymethylene-3-oxo-3-(polyfluoroalkyl) propionates with 3-amino-5-hydroxypyrazole result in ethyl 2,7-dihydroxy-7- (polyfluoroalkyl)-4,7-dihydropyrazolo[1,5-a]pyrimidin-6-carboxylates under mild conditions.
