Аннотация: The data on the chemistry of fluorinated quinolines available in the literature of the last 10–15 years are presented. A variety of synthetic methods exploiting cyclization and cycloaddition reactions, displacements of halogen atoms or the diaza group, as well as direct fluorinations have been considered. Novel approaches to functionalization of polyfluorinated quinolines, including nucleophilic displacement of fluorine atoms, cross-coupling reactions, and synthesis on the basis of organometallic compounds are discussed. Selected representative examples of fluoroquinolines exhibiting a remarkable biological activity or those quinolines which have already found their applications in medicine will also be discussed in the text. Аннотация: Tri- and pentacyclic fluoroquinolones were synthesized by intramolecular cyclization of the corresponding 3-hydrazinopolyfluorobenzoylacrylates followed by substitution of fluorine atoms by amine residues. The antitumor activity of the resulting compounds was studied at the National Cancer Institute using cultures of 60 cell lines of nine groups, including leukemia, lung tumor, large intestine tumor, CNS tumor, melanoma, ovary tumor, renal tumor, prostate tumor, and breast tumor. Relationships between structure and antitumor activity were analyzed. In vivo experimental data from hollow fiber tests are presented for two derivatives

Аннотация: 2-Methylthio-4-phenylamino-6,7,8-trifluoro-3H-quinazolin-4-one was synthesized by means of intramolecular cyclizations of S-methyl-N-(tetrafluorobenzoyl)isothiourea followed by a reaction with POCl3 and nucleophilic substitution at the 4-position. The reactions of the synthesized compound with amines proceed, depending on their nature, via substitution of either the F(7) atom or the SMe fragment in the 2-position. The antiviral activity of the obtained 6,7,8-trifluoro-2-ethylthioquinazolin-4-ones was investigated using monkeypox virus, smallpox vaccine, and ectromelia virus. It is shown that fluorinated quinazoline derivatives have good prospects in the search for new active substances

Аннотация: A new fluorine-containing tetradentate ligand 1,2-bis(5,5,5-trifluoro-4-oxopent-2-en-2-amino)benzene and its complexes with Ni(II), Pd(II) and Cu(II) are characterized by single crystal X-ray diffraction. It is found that the enaminoketone fragments of the ligand are identical in bond lengths and angles; they are almost planar, and make the angles of 51.3° to the plane of the benzene ring. The structures of Ni(II), Pd(II), and Cu(II) complexes are similar and have a saddle-shape configuration. The metal ions have square planar coordination and are located almost in the center of the N2O2 square. The average M-N bond lengths are longer than M-O ones by 0.014 A and 0.034 A for the Ni(II) and Cu(II) complexes respectively, while in the Pd(II) complex, M-O is longer than M-N by 0.029 A. The average chelate angles N-M-O in the complexes are: N-Ni-O 95.12°; N-Pd-O 95.68°; N-Cu-O 93.88

Аннотация: New synthetic approaches to fluorinated 3-phenyl-1,2,4-benzotriazines for biological testing have been elaborated. 1-(3,4-Difluorophenyl)-3,5- diphenylformazan (IVa) was synthesized via dinitriding of 3,4-difluoroaniline, followed by azo-addition of the resulting azobenzenediazonium chloride with acetaldehyde phenylhydrazone. 6,7-Difluoro-3-phenyl-1,2,4-benzotriazine (Va) was obtained via intramolecular cyclization of formazan IVa in the presence of BF3/AcOH complex. Monofluoro-substituted 6-R-7-fluoro-3-phenyl-1,2,4- benzotriazine derivatives (Vb-Vq) were prepared by substituting fluorine atom with alkoxides in 3,4-difluoronitrobenzene. Conditions for nucleophilic substitution of the second fluorine atom in benzotriazines V have been established. Fluorinated 3-phenyl-1,2,4-benzotriazines have been tested for antiviral and cytotoxic activity on Vero cell cultures and proved to be active against severe diseases caused by smallpox and some other pathogenic viruses

Аннотация: A series of 5,6-disubstituted benzimidazole nucleosides, obtained earlier, did not show any significant antiviral activity at relatively low cytotoxicity in vitro. In the course of our research we have succeeded in introducing an additional fluorine atom into the benzimidazole ring system. A new series of 4,6-difluorobenzimidazoles, bearing various groups (fluoro-, methoxy-, ethoxy-, morpholino-, and pyrrolidino-) in the 5-position of the benzene ring, have been synthesized. All these compounds proved to be substrates for recombinant E. coli purine nucleoside phosphorylase (PNP) in the transglycosylation reaction. Effective methods for the synthesis of ribo- and 2′-deoxyribonucleosides with high yields (60–90%) have been described, and the formation of regioisomeric N3-nucleosides of benzimidazoles have been detected. The biological activity of the nucleosides obtained against herpes simplex virus type 1 (HSV-1) has been elucidated. All compounds show a low cytotoxicity in the cell culture Vero E6. 4,5,6-Trifluoro-1-(β-d-ribofuranosyl)benzimidazole and 5-methoxy-4,6-difluoro-1-(β-d-2′-deoxyribofuranosyl)benzimidazole proved to inhibit completely the progression of the virus cytopathic effect (CPE) at a multiplicity of infection (MOI) of 0.01 PFU/cell.

Аннотация: Novel N,O-bidentate BF2 and BPh2 complexes have been prepared in good to excellent yields through coordination of 8-hydroxy-2-methylquinolines and 2-(2-hydroxyphenyl)-3H-quinazolin-4-ones with boron trifluoride etherate or triphenylborane under mild conditions. All complexes have been characterized by H-1, B-11 and F-19 NMR, mass-spectrometry and X-ray crystallography data. Some complexes have been found to exhibit a significant fluore.scence in acetonitrile solutions. Electronic and site effects of substituents in both heterocyclic and phenol fragments proved to have a profound impact on quantum yields.
