The features of tautomerism in the series of asymmetrical 5(6)-fluorobenzofuroxans in acetone solutions have been studied by 1H, 13C and 19F NMR.????

Аннотация: New synthetic routes to trifluoromethylated N-heterocycles based on condensations of 2-methoxy-2-methyl-5-(trifluoromethyl)furan-3(2H)-one with bifunctional N-nucleophiles are described. For the first time, trifluoromethyl-containing pyrazoles, pyrazolines and isoxazolines bearing hydrazone or oxime groups could be obtained by a one-pot strategy based on reactions of 5-(trifluoromethyl)furan-3(2H)-one with two equivalents of the corresponding hydrazines or hydroxylamine in the presence of an acid. The interaction of furan-3(2H)-one with ureas proceeded under mild conditions to furnish 1H-furo[2,3-d]imidazol-2(3H)-one derivatives in good yield. Further, a trifluoromethyl-containing quinoxaline derivative was obtained by condensation of furan-3(2H)-one with ortho-phenylenediamine.

Аннотация: A new approach for the synthesis of fluorinated 1H-quinazolin-4-ones and 4-substituted quinazolines has been developed. 6-Fluoro-1H-quinazolin-4-ones were obtained by intramolecular cyclization of fluorine-containing S-ethyl N-benzoylisothioureas. Nucleophilic substitution reactions at positions 2 and 7, as well as alkylation at 1-position of quinazolinones were investigated. In addition, the synthesis of fluorine-containing 4-aminoquinazolines was carried out.

Strong emphasis is put on metalloproteins, which play a significant role in the biochemistry of vitally essential processes, as well as metal compounds that are widely used in medicine. Of particular importance are the chemical elements and the isotopes, compounds of which are employed in nuclear medicine for diagnostics and treatment of a wide range of diseases, oncological and cardiovascular in particular.

Аннотация: A series of 5,6-disubstituted benzimidazole nucleosides, obtained earlier, did not show any significant antiviral activity at relatively low cytotoxicity in vitro. In the course of our research we have succeeded in introducing an additional fluorine atom into the benzimidazole ring system. A new series of 4,6-difluorobenzimidazoles, bearing various groups (fluoro-, methoxy-, ethoxy-, morpholino-, and pyrrolidino-) in the 5-position of the benzene ring, have been synthesized. All these compounds proved to be substrates for recombinant E. coli purine nucleoside phosphorylase (PNP) in the transglycosylation reaction. Effective methods for the synthesis of ribo- and 2′-deoxyribonucleosides with high yields (60–90%) have been described, and the formation of regioisomeric N3-nucleosides of benzimidazoles have been detected. The biological activity of the nucleosides obtained against herpes simplex virus type 1 (HSV-1) has been elucidated. All compounds show a low cytotoxicity in the cell culture Vero E6. 4,5,6-Trifluoro-1-(β-d-ribofuranosyl)benzimidazole and 5-methoxy-4,6-difluoro-1-(β-d-2′-deoxyribofuranosyl)benzimidazole proved to inhibit completely the progression of the virus cytopathic effect (CPE) at a multiplicity of infection (MOI) of 0.01 PFU/cell.

Аннотация: An efficient approach for the synthesis of 3-(polyfluoroacyl)pyruvaldehydes dimethyl acetals (1,1-dimethoxy-4-polyfluoroalkyl-butan-2,4-dions) from 1,1-dimethoxyacetone and polyfluorinated carboxylic acid esters has been developed. The procedure includes the Claisen type condensation of the starting materials by means of calcium hydride in methanol, followed by isolation of copper complexes of the corresponding diketones and their destroying with disodium EDTA. A simple synthesis of 5(3)-(polyfluoroalkyl)-1H-pyrazole-3(5)-carbaldehydes and their acetals is also presented.

Аннотация: Fluorinated enaminoketones, a new type of ligands, bearing two independent coordination centers (polyether's and fluorinated enaminoketones fragments) have been obtained. The crystal structure of the Cu(II) complex of 1,5-bis-[2-(4`,4`,4`-trifluoro-1`-methyl-3`oxo-but-1`-enylamino)-phenoxy]-3-oxapentane (10) has been elucidated by X-ray crystallography. The results obtained show that the complex 10 consists of two crystallographically independent molecules C26H24CuF6N2O5 (A and B), and the metal atom in the complex 10 has four-coordinated arrangement, as a polyhedron with a distorted square with two nitrogen and two oxygen atoms located in corners.

Аннотация: The data on the chemistry of fluorinated quinolines available in the literature of the last 10–15 years are presented. A variety of synthetic methods exploiting cyclization and cycloaddition reactions, displacements of halogen atoms or the diaza group, as well as direct fluorinations have been considered. Novel approaches to functionalization of polyfluorinated quinolines, including nucleophilic displacement of fluorine atoms, cross-coupling reactions, and synthesis on the basis of organometallic compounds are discussed. Selected representative examples of fluoroquinolines exhibiting a remarkable biological activity or those quinolines which have already found their applications in medicine will also be discussed in the text.
